Annealases are ATP-independent single-strand DNA (ssDNA) binding proteins that catalyze annealing of a ssDNA to a homologous ssDNA to form a double-strand DNA (dsDNA).[1]

Most annealases are DNA recombination proteins, though some may have additional activities resulting in multi-functionality. Most, if not all, annealases found in dsDNA viruses form a complex with its partner viral exonuclease to form a DNA recombinase referred to as an EATR complex (Exonuclease Annealase Two-component Recombinase). EATR complexes act as recombinases that catalyze a ssDNA annealing homologous DNA recombination reaction, which is commonly referred to as a single-strand annealing (SSA) homologous recombination.

Annealases are different from the ATP-dependent strand invasion proteins such as the E. coli RecA DNA recombination protein. Redβ annealase of bacteriophage lambda (phage λ) is most likely the first annealase identified.
