The highway starts in the west at a junction with Fed37 about {{convert|10.6|km|mi|abbr=on}} south-southwest of Manuel Doblado, Guanajuato. The highway travels east-northeast for {{convert|30|km|mi|abbr=on}} before heading mostly south for {{convert|11.7|km|mi|abbr=on}} toward Cuerámaro. From Cuerámaro, FH 41 travels {{convert|28|km|mi|abbr=on}} east-southeast until reaching its eastern terminus at Fed. 90 in the locale of Munguia, Guanajuato.[2] The highway's eastern terminus is {{convert|12.5|km|mi|abbr=on}} southwest of Irapuato, Guanajuato.
